Depending on the supplier and test package, a peptide COA may include: the supplier name and address; the product name, sequence, and CAS number where applicable; the batch (lot) number and manufacture date; purity testing data with supporting chromatogram where available; appearance notes; solubility notes; water content when relevant; residual solvents; counter-ion content; and the signature of the analyst or quality control officer. Some suppliers add extra identity or biological-assay tests when those tests have been run for the relevant batch. The COA is important for traceability in published research: when you cite 'Peptide X (Peptra Labs, batch 12345)' in a publication, the batch number lets other researchers - or peer reviewers - retrieve the exact batch record where available. Without batch documentation, batch-to-batch reproducibility cannot be claimed. Peptra Labs keeps batch-level CoA records where available; if you need to check a specific lot for an audit, publication, or institutional procurement, email info@peptralabs.com with the batch number from the vial label. ### HPLC purity *Also known as: HPLC purity grade, high-performance liquid chromatography* Source: https://www.peptralabs.ro/glossary/hplc-purity High-performance liquid chromatography (HPLC) is the industry standard for measuring peptide purity. When a peptide has a stated HPLC purity percentage, that figure should come from a calibrated HPLC system, typically using a reverse-phase C18 column with a UV detector at 214 nm or 220 nm. Each peak in the chromatogram represents a distinct molecular species; the area under the target peak, expressed as a percentage of the total peak area, is the purity figure. For research peptides, very high purity values should still be reviewed alongside any extra batch tests that were actually run, without treating purity as proof of molecular identity. The choice of UV detection wavelength matters: 214 nm captures the peptide bond absorption directly and is the most universal setting for peptides; 220 nm is sometimes used to suppress mobile-phase noise but underestimates absorbance for peptides without aromatic residues. Gradient HPLC (varying mobile phase composition over time) separates closely related species - truncated sequences missing one residue, oxidized variants, racemized residues - that would co-elute under isocratic conditions. The full purity report on a Certificate of Analysis should include the column type, mobile phase, gradient program, flow rate, detection wavelength, retention time of the target peak, and a copy of the chromatogram itself. Reviewing all of these is the only way to understand what a stated purity figure means for the relevant batch. Peptra Labs records these parameters in available COA information for eligible batches. ### Lyophilised (freeze-dried) *Also known as: freeze-dried, lyo* Source: https://www.peptralabs.ro/glossary/lyophilised Lyophilisation, also called freeze-drying, is the standard preservation method for peptides intended for long-term storage and shipping. The process involves freezing the peptide solution rapidly to lock the molecular structure in place, then removing the water by sublimation under high vacuum at low temperature. The result is a fluffy white to off-white solid - the lyophilizate - that contains the original peptide molecules in a dry, glassy state without thermal degradation. Lyophilised peptides offer several practical advantages for research supply: dramatically extended shelf life (years at -20°C versus days to weeks for aqueous solutions); resistance to microbial contamination (water is required for microbial growth); preserved chemical integrity (no hydrolysis, oxidation, or deamidation in the dry state); and reduced shipping cost and complexity (no cold chain required for transit). The lyophilizate is typically reconstituted at the lab bench just before use by adding sterile water, bacteriostatic water, or a specific buffer to the vial, with gentle inversion to dissolve. The mass shown on the COA refers to the net peptide content, not the total lyophilizate weight (which may include counter-ions, residual buffer salts, or excipients). For peptides supplied with bulking agents like mannitol, the COA should specify both the peptide content and the total fill weight. Storage of the unreconstituted lyophilised vial at -20°C in a sealed container is the gold standard; some labs use -80°C for extra long-term storage. Once reconstituted, the solution should be aliquoted and stored according to peptide-specific guidance to avoid freeze-thaw degradation. ### Mass spectrometry (MS) *Also known as: MS* Source: https://www.peptralabs.ro/glossary/mass-spectrometry Mass spectrometry (MS) is an analytical technique used to evaluate molecular mass by measuring mass-to-charge ratio. While HPLC measures purity (how much of one species there is), MS can add molecular-mass context for the species being reviewed. For peptides, the typical workflow uses electrospray ionization (ESI) or matrix-assisted laser desorption ionization (MALDI) to produce charged molecular ions, which are then separated and detected by a mass analyzer (quadrupole, time-of-flight, or orbitrap). The observed monoisotopic mass is compared to the theoretical mass calculated from the peptide sequence; a close match can support batch review when that test is available. Such testing can complement the HPLC purity number, but it should not be assumed for a batch unless the relevant CoA or lab record shows it. A peptide can be highly pure by HPLC while still requiring additional review for molecular identity if the research protocol depends on that distinction. For peptides with complex modifications - phosphorylation, biotinylation, fluorescent tags, cyclization - tandem MS (MS/MS) may be used to fragment the peptide and review sequence coverage. Peptra Labs only presents MS characterization for batches where that testing exists in the relevant record. ### Peptide *Also known as: polypeptide, amino acid chain, oligopeptide* Source: https://www.peptralabs.ro/glossary/peptide A peptide is a molecule formed when two or more amino acids are joined by covalent peptide bonds - the same amide linkages that build proteins. The defining feature of a peptide versus a protein is length: peptides are typically short, ranging from two amino acids (a dipeptide) to roughly fifty residues. Beyond that threshold the molecule is conventionally classified as a polypeptide or a protein, although the boundary is set by convention rather than chemistry. Peptides are described by their amino acid sequence, written in single-letter or three-letter code from the N-terminus (free amino group) to the C-terminus (free carboxyl group). Each residue contributes a side chain whose physical and chemical properties - charge, polarity, hydrophobicity, hydrogen-bonding capacity - determine how the peptide folds and how it interacts with other molecules. Peptides are produced biologically by ribosomal translation or non-ribosomal peptide synthetases, and synthetically by solid-phase peptide synthesis (SPPS), the method that underpins almost all research-grade material today. In a research setting, peptides are valuable because they sit at a molecular size where they can be both highly specific in binding and chemically defined enough for reproducible study, which is why so much biochemical and biophysical research is built around them. Peptra Labs supplies these peptides for laboratory research use only, with available batch-level CoA information focused on purity testing where applicable. ### Peptide storage *Also known as: peptide stability* Source: https://www.peptralabs.ro/glossary/storage Proper storage is critical to preserving peptide purity, identity, and biological activity over time. The standard recommendation for lyophilised research peptides is -20°C in a sealed glass vial, protected from light and moisture, with a desiccant in the storage container. Under these conditions, most peptides are stable for years (often 2-5 years, with stability data available on COA where measured). For peptides containing residues prone to degradation - cysteine (oxidation of the free thiol), methionine (oxidation to sulfoxide), tryptophan (photo-oxidation), asparagine and glutamine (deamidation), N-terminal glutamine (cyclization to pyroglutamate) - colder storage at -80°C and stricter exclusion of oxygen and light extend shelf life further. Reconstituted aqueous solutions are far less stable than the lyophilised form. The general rule for reconstituted peptides is: use within 28 days at 2-8°C if bacteriostatic water was used, or within 24-48 hours if sterile water without preservative. For long-term storage of reconstituted material, aliquot into single-use volumes and store at -20°C or -80°C; avoid freeze-thaw cycles which cause physical aggregation and chemical degradation. The thumb rule: each freeze-thaw cycle typically reduces peptide integrity by 5-10% depending on the peptide's intrinsic stability. Document storage conditions in the lab notebook - many published papers fail reproducibility audits because storage history is unclear. Done correctly, it preserves the peptide's purity, identity, and biological activity; done carelessly, it can introduce contamination, partial degradation, or inconsistent concentrations across replicate experiments. The choice of solvent depends on the peptide and the downstream application. Sterile water (USP grade) is the simplest choice for short-term reconstitution and gives a clean, unbuffered solution. Bacteriostatic water (containing 0.9% benzyl alcohol) inhibits microbial growth and is suitable for multi-use vials over 1-28 days at 2-8°C, depending on the peptide's intrinsic stability. Specific buffers - PBS at pH 7.4, acetate at pH 4-5, ammonium bicarbonate for downstream MS - are chosen when the peptide's pI, solubility, or experimental context requires it. Some hydrophobic peptides require an initial dissolution in a small volume of DMSO or acetic acid before dilution into the working buffer. The volume to add is calculated to give the desired final concentration: dividing the labelled mass on the vial by the volume yields the concentration. Most labs aliquot the reconstituted solution into single-use volumes immediately, store the aliquots at -20°C or -80°C, and thaw one aliquot per experiment to avoid freeze-thaw degradation of the bulk stock. Document the reconstitution conditions in the lab notebook: solvent, volume, date, expected concentration, and any visual observations (cloudiness, precipitate, color). This documentation is essential for reproducibility and is often requested by reviewers when reporting peptide-based research. ### Research Use Only (RUO) *Also known as: for research use only, non-clinical research* Source: https://www.peptralabs.ro/glossary/ruo RUO stands for 'Research Use Only' - a regulatory designation that marks a compound, kit, or material as intended exclusively for in-vitro or laboratory research, NOT for diagnostic, therapeutic, or human consumption purposes. The RUO label is recognized in major regulatory frameworks (FDA in the US, IVDR in the EU, similar regimes elsewhere) and signals that the supplier has neither sought nor obtained approval for the product as a drug or medical device. RUO products may be characterized with research-grade purity data, but they have not gone through clinical trials, GMP manufacturing, sterility validation for parenteral use, or biocompatibility testing - all of which are required before a substance can be sold for human therapeutic use. The RUO designation has important practical consequences: research peptides cannot legally be marketed with health claims; they cannot be prescribed; they cannot be imported by individuals in many jurisdictions for personal use without an institutional research license. Within a properly licensed research laboratory, they are tools for studying biological mechanisms in cell cultures, model organisms, or biochemical assays. Some research peptides have advanced into clinical trials and become approved drugs (e.g., the GLP-1 analog semaglutide), but the RUO version supplied for laboratory research is chemically the same compound under a different regulatory category.
